The purpose:

Demonstrate how the ELISA assay is used to diagnose exposure to a disease by testing for the presence of antibodies to the disease in a sample of simulated serum.
Determine the hemagglutinating antibody titer of two different sera.
Demonstrate Complement Mediated Lysis and the sequence of reactions necessary to cause lysis.

Student bench setup

  1. Rack 1: 5 x empty 15ml tubes for Complement Lysis, 2 x 10ml tubes with saline, 1 x 10ml tube with 1%SRBC
  2. Rack 2: Complement Lysis reagents
  3. Rack 3: Elisa reagents
  4. Biorad 12 well strip
  5. Stack of brown paper
  6. 96 U-bottom well plate and towels seal for hemagglutination
  7. Wash buffer with plastic 1 ml transfer pipette
  8. Liquid Waste container
  9. Bio-hazard waste
  10. Timer
  11. 5 ml serological pipette and Pi-pump
  12. 2x Markers (fine tip)
  13. Pasteur pipettes and bulbs
  14. Adjustable 1000 µl and 200 µl pipettes and tips
  15. Centrifuge

  1. Water bath at 37 °C + plastic racks
  2. Tray to place hemagglutination plates going to Incubator
  3. Bio-hazard waste
  4. 200 µl pipette tips
  5. Serum 1
