Safe use of a centrifuge
Centrifugation (Video transcript)
A centrifuge applies centripetal force to a sample to allow for separation of its different components. These operate at high speed and it is important to follow several safety precautions.
First, make sure the centrifuge is not in use before you open the lid. You can test this by placing your hand on top of the centrifuge to feel for vibrations.
STEP 1: it is important to load the samples properly.
The samples must be balanced for position and weight. GENERALLY SPEAKING, the larger the centrifuge the more critical it will be that the sample weight is balanced due to the higher forces being generated.
The central component of the centrifuge is called the rotor and it is where the samples are held. Depending on the type of rotor you have will determine how you must balance your tubes.
For some types of rotors a symmetrical arrangement would be okay. BUT NOT OUR centrifuge.
For this type of rotor we must place the tubes across from one another. *Any time you use a new centrifuge, be sure that you understand how it works *
Because we have an odd number of samples, we need to include a balance tube – that contains a similar amount of solution.
Once the tubes are loaded in a balanced fashion, close and lock the lid.
Select the desired speed.
We will use our centrifuge at its maximal setting.
Set the timer and spin the sample for the desired amount of time.
When the spin is complete, turn the centrifuge to the off position.
Be sure to wait until the centrifuge has stopped before unlocking and opening the lid and removing your samples.
